Effect of Concomitant Administration of Aqueous Extract of Pleurotus pulmonarius and Metformin in Diabetic Mice
Abstract
The present investigation studied the interaction of the aqueous extract of Pleurotus pulmonarius (Fr.) Quel.-Champ (Lentinacae) (called PP-aqu) with metformin on serum glucose level, body weight and oral glucose tolerance test (OGTT) in alloxan-induced diabetes in mice. PP-aqu (500 mg/kg), metformin (250 and 500 mg/kg) singly and concomitantly were administered orally in alloxan-induced (70 mg/kg, i.v.) diabetic mice. In the acute study, serum glucose levels were determined at 0, 2, 4, 6 and 24 h after drug administration. The sub-acute study involved repeated administration of drugs for 14 days. In OGTT, D-glucose (2.5 g/kg) was administered in diabetic mice 30 min after pretreatment with PP-aqu (500 mg/kg), metformin (250 and 500 mg/kg) and their concomitant administration. The onset and peak antihyperglycaemic effect of PP-aqu (500 mg/kg) and metformin (250, 500 mg/kg) alone was 2 h. Onset of concomitant administration of PP-aqu (500 mg/kg) with metformin (250 and 500 mg/kg) was 2 h; the peak effect was 6 h and the duration of action was 24 h. Repeated administration (once a day for 14 days) of metformin, PP-aqu and their concomitant administration caused significant (P<0.001) reduction in the serum glucose level as compared to vehicle treated group. Concomitant treatment prevented decrease in body weight of diabetic mice. In OGTT, concomitant administration of PP-aqu with metformin increased glucose threshold. The concomitant administration of PP-aqu with metformin produced synergistic antihyperglycaemic effect than either drug alone.
